Yang Yunnan Provincial Infectious Disease Hospital, Kunming, China General data Abstract category: Antiretroviral therapy – observational studies Abstract body Purpose: DTG/3TC Phase III studies have demonstrated non-inferiority of DTG + 3TC versus a variety of 3/4DRs in virologically suppressed participants (TAF-based: TANGO and CAR: SALSA). We conducted this analysis to better understand real-world use of DTG/3TC in virologically suppressed China PLHIV. Method: This study was China-based, retrospective chart review. PLHIV were selected for inclusion based on the following criteria: age ≥18 years, using DTG+3TC for any reason between January 2021and March 2022. The primary objective was the rate of virological suppression( defined as an HIV-1 VL less than 50 copies/ml). Changes in laboratory parameters were also evaluated. Results: 121 patients who met study criteria were eligible for the analysis. Overall, mean age was 47.5 (25.5-73.5) years and 84(66.1%) of the cohort were male. The most common reason for switching to the dual therapy was adverse effects(59.5%), request for simplification (36.4%), followed by renal toxicity (32%) and hyperlipidemia(15.3%). At 48 weeks, the proportion of patients maintaining virological suppress were 99.17%. Four patients with archived M184V/I receiving DTG + 3TC and zero VF events are observed. We found a increase in CD4+ counts at 48 weeks after switching drug strategies(726 vs. 755, p>0.05).And we also detected a signaficant increase in the CD4+/CD8+ count rate at 48 weeks(0.763 vs. 0.819, p<0.05). We haven’t detected any significantly change in TG, TC and HDL-C between baseline and 48 weeks. DTG+3TC regimen had increased LDL-C (2.66mmol/L vs.2.82mmol/L, p<0.05) and decreased eGFR( 90.99 mL/min·1.73 m vs. 99.12mL/min·1.73 m ,p<0.05) Weight gain averaged 1.09 kg(p<0.05) with DTG+3TC. Conclusions: PLHIV who, being virologically suppressed, switched to dual therapy based on DTG/3TC, experienced a statistically significant increase in CD4/CD8 T cell ratio, as well as high efficacy in terms of viral suppression.
