Pacheco Hospital Prof. Doutor Fernando Fonseca, Infectious Diseases, Lisbon, Portugal General data Abstract category: Antiretroviral therapy – observational studies Abstract body Purpose: To assess the effectiveness of tenofovir alafenamide (TAF)/emtricitabine (FTC)/bictegravir (BIC) in people with HIV-2 (PWHIV-2) in a real-world setting. Method: Retrospective cohort of PWHIV-2 on TAF/FTC/BIC since November 2019 in an HIV clinic in Portugal. Virological and immunological variables were evaluated and compared between prior to TAF/FTC/BIC initiation and month 12 (M12). Results: Thirty-seven patients were included, with a median age of 56 years (48-68), mostly female (73%, 27/37) and from West Africa (95%, 35/37). Among 9 treatment-naïve PWHIV-2, median baseline CD4 count, CD4/CD8 ratio and plasma viral load (pVL) were respectively 467 cells/µL (212-728), 0,45 (0,27-0,91) and 146 copies/mL (58-1264). At M12, all patients had pVL<40 copies/mL (p=0,016). Twenty-eight treatment-experienced PWHIV-2 were included. Median CD4 count and CD4/CD8 ratio were 583 cells/µL (343-683) and 0,83 (0,44-1,15). Prior to TAF/FTC/BIC 71% of patients were virologically suppressed. Most non-suppressed patients (6/8, 75%) had prolonged treatment interruption (up to 36 months). Prior drug regimens were based on integrase (80%) or protease (20%) inhibitors. Main reasons for switch were treatment simplification (17/25, 68%) and minimising toxicity (11/25, 44%). TAF/FTC/BIC was discontinued in one patient due to declining CD4 count at month 1. At M12, 26/27 patients (96%) had pVL<40 copies/mL (p=0,016). Median CD4 count and CD4/CD8 ratio were 566 (390-746) cells/µL and 0,89 (0,47-1,32) respectively (p=0,249 and p=0,01). No major adverse events or opportunistic diseases were reported. Conclusions: This is the first cohort of TAF/FTC/BIC use in PWHIV-2. Our data demonstrated good efficacy and safety.
